Effects of Heat Shock Pretreatment on the Lipoplysaccharid-Induced Macrophage Migration

    Abstract:

    Aim To study the effects of heat shock pretreatment on the lipopolysaccharid-induced macrophage migration. Methods The effect of lipopolysaccharid on the release of tumor necrosis factor-α(TNF-α) and interleukin-1β(IL-1β) in murine macrophage-like RAW264.7 cells were detected by ELISA;the effect of lipopolysaccharid on the migration of RAW264.7 macrophages were detected by chemotaxis assay;the effect of heat shock pretreatment on the migration of RAW264.7 macrophages were detected by chemotaxis assay. Results Interfered with 500 μg/L lipopolysaccharid for 1 h,the release of TNF-α and IL-1β were significantly increased(P<0.05) in RAW264.7 macrophages;interfered with 500 μg/L lipopolysaccharid for 24 h,the migration of RAW264.7 macrophages was significantly increased(P<0.05);given a heat shock pretreatment about 42.5℃ for 1 h followed with the interference of 500 μg/L lipopolysaccharid,the migration of RAW264.7 macrophages induced by lipopolysaccharid was significantly inhibited(P<0.05). Conclusion Heat shock pretreatment significantly inhibited lipopolysaccharid-induced macrophage migration.
