    摘要:

    目的 探讨辣椒素对大鼠肾缺血再灌注损伤的保护作用及线粒体相关作用机制。方法 将50只雄性SD大鼠分成假手术组、肾缺血再灌注损伤组和辣椒素低、中、高剂量组。采用夹闭双侧肾蒂构建肾缺血再灌注损伤模型。肾缺血45 min,再灌注24 h,过量麻醉法处死大鼠,收集肾脏和血清。检测血清肌酐(SCr)、血尿素氮(BUN)、肾脏组织病理形态和细胞凋亡,测定线粒体三磷酸腺苷(ATP)和丙二醛(MDA)含量以及Ca2+-ATP 酶、Na+-K+-ATP酶、过氧化氢酶(CAT)、谷胱甘肽过氧化物酶(GPx)和超氧化物歧化酶(SOD)活性。结果 辣椒素干预可减少SCr和BUN含量,降低肾组织病理改变和细胞凋亡,增加线粒体Ca2+-ATP酶、Na+-K+-ATP酶、CAT、GPx和SOD酶活性以及ATP的含量,但减少MDA的水平。结论 辣椒素对肾缺血再灌注损伤有保护作用,其作用呈浓度效应,机制与抑制线粒体脂质过氧化相关。

    Abstract:

    Aim To investigate the protective effect of capsaicin on renal ischemia reperfusion injury in rats and its mechanism. Methods 50 male SD rats were divided into sham operation group (Sham), renal ischemia-reperfusion injury group (IRI) and low, middle and high dose capsaicin group. The model of renal ischemia reperfusion injury was established by clamping bilateral renal pedicle for ischemia 45 min, and reperfusion 24 h. Then rats were killed by excessive anesthesia and kidney and serum were collected. Serum creatinine (SCr), urea nitrogen (BUN), cell apoptosis and pathologic renal tissue were detected, mitochondrial adenosine triphosphate (ATP), malondialdehyde (MDA) content, Ca2+-ATP enzyme,Na+-K+-ATP enzyme, catalase (CAT), glutathione peroxidase (GPx) and superoxide dismutase (SOD) activity were measured. Results Capsaicin intervention can reduce BUN, SCr, cell apoptosis and the pathological change of kidney tissue, increase Ca2+-ATP, Na+-K+-ATP, CAT, GPx and SOD enzyme activity and ATP content and decrease the level of MDA in mitochondria. Conclusion Capsaicin has protective effects on renal ischemia-reperfusion injury in a concentration dependent-manner and its mechanism is related to the inhibition of lipid peroxidation.
