    摘要:

    目的 探讨桂北地区汉族代谢综合征(MS)与脂联素启动子单核苷酸多态性(SNP)的相关性。方法 筛选108例MS患者为MS组和76名健康者为对照组,测量身高、体重、血压、空腹血糖(FPG)、甘油三酯(TG)、总胆固醇(TC)、低密度脂蛋白胆固醇(LDLC)、高密度脂蛋白胆固醇(HDLC)、丙氨酸氨基转移酶(ALT)、天门冬氨酸氨基转移酶(AST)、尿酸(UA)以及进行肝脏B超检查,比较两组人群临床资料的差异,并对MS相关危险因素进行非条件Logistic回归分析。同时应用聚合酶链反应-限制性内切酶片段长度多态性(PCR-RFLP)技术,分析两组人群脂联素基因启动子-11377C/G位点基因型和等位基因频率差异,以及不同基因型相关体格测量和生化指标的差异。结果 MS组和对照组之间临床资料(体质指数、血压、FPG、TG、TC、LDLC、HDLC、ALT、AST和UA)比较,差异有统计学意义(P<0.01或P<0.05);Logistic回归分析结果显示,性别、年龄、尿酸以及脂肪肝是MS的独立危险因素(P<0.01或P<0.05);脂联素基因启动子-11377C/G位点在MS组与对照组之间比较基因型和等位基因频率差异均无统计学意义;MS组和对照组脂联素基因启动子-11377C/G不同基因型体格测量和生化指标比较,差异均无统计学意义。结论 脂联素基因启动子SNP-11377C/G与桂北地区汉族MS的发病无明显相关性。

    Abstract:

    Aim To investigate the association of adiponectin promoter single nucleotide polymorphisms with metabolic syndrome in Hans of the northern Guangxi. Methods A total of 108 MS subjects were selected as MS group, and 76 normal adults were selected as control group. Measurement of height, weight, blood pressure, fasting blood glucose(FPG), triglyceride (TG),total cholesterol(TC),low density lipoprotein cholesterol(LDLC),high density lipoprotein cholesterol (HDLC),uric acid(UA) and ultrasonic examination were performed and these clinical datas were compared between the two groups. The attributing factors of MS were analyzed by Logistic regression model. The polymerase chain reaction-restriction fragment length polymorphism(PCR-RFLP) was applied to analyze the differences of adiponectin gene promoter -11377C/G in the genotype and allele frequency between cases and controls. Many anthropometric and biological indices were compared between cases and controls in different genotype carriers. Results There was significant difference of clinical data(body mass index,biood pressure,FPG,TG,TC,LDLC, HDLC and UA) between MS and control groups(P<0.01 or P<0.05). The multivariable stepwise analysis extracted sex,age,UA and fatty liver as major risk factors for MS(P<0.01 or P<0.05). There was no significant difference of the distribution of the genotype and allele frequencies between MS and control groups. The anthropometric and biological indices were not significant among different genotypes in MS and control groups. Conclusions The adiponectin gene promoter -11377 is not associated with metabolic syndrome in Hans of the northern Guangxi.
