    摘要:

    目的观察和比较高脂与糖尿病小鼠脑组织中血红素加氧酶1和血红素加氧酶2的表达水平。方法应用逆转录聚合酶链反应、Western Blotting和免疫组织化学技术分别检测高脂饲喂、高脂饲喂+链脲佐菌素诱导的糖尿病小鼠脑组织血红素加氧酶1和血红素加氧酶2的mRNA及蛋白的表达。结果高脂组和糖尿病组血红素加氧酶1的mRNA和蛋白表达均明显升高,mRNA表达水平分别是相应对照组的1.63倍和1.60倍(P均<0.01);但血红素加氧酶2表达仅在糖尿病组升高(P><0.01),其蛋白表达水平为对照组的1.83倍(P><0.01)。结论糖尿病小鼠脑组织血红素加氧酶系统高水平表达。

    Abstract:

    Aim To observe and compare the expression of heme oxygenase-1 and-2 in the brain of high fat and diabetic mouse.Methods Heme oxygenase expression was detected by reverse transcription polymerase chain reaction(RT-PCR),Western Blotting and immunochemistry.Results More significant heme oxygenase-1 mRNA and protein expression were observed both in the brain of high fat mouse and diabetic mouse,and the mRNA expression were 1.63 times and 1.60 times of those of control group.Elevated expression of heme oxygenase-2 were observed only in the brain of diabetic mouse,and the quantitative evaluation showed that heme oxygenase-2 protein was increased 1.83-fold in the diabetic group compared with that of control group.Conclusion Heme oxygenase was over-expressed in the brain of diabetic mouse.
