    摘要:

    目的观察糖基化终末产物对大鼠血管平滑肌细胞钠/氢交换蛋白1活性的影响。方法采用组织贴块法原代培养大鼠胸主动脉平滑肌细胞。用体外制备的外源性糖基化终末产物和血管平滑肌细胞共孵育24h,利用BCECF通过荧光分光光度计检测细胞上钠/氢交换蛋白1活性。结果不同浓度糖基化终末产物(1、5和10mg/L)与血管平滑肌细胞共孵24h后,能显著增强钠/氢交换蛋白1的活性,分别使钠/氢交换蛋白1的活性从0.66±0.05升高至0.71±0.03、0.80±0.07和0.88±0.06(P<0.05或P<0.01),使钠/氢交换蛋白1 mRNA表达从0.77±0.07分别升高至0.88±0.08、1.23±0.06和1.33±0.08(P<0.05或P<0.01);预先使用糖基化终末产物受体阻断剂5 mg/L,钠/氢交换蛋白1活性比糖基化终末产物处理组明显降低(0.68±0.04比0.90±0.05,P<0.05);使用不同浓度的丝裂原活化蛋白激酶阻断剂PD98059(1、10和25μmol/L),使钠/氢交换蛋白1活性从0.94±0.05分别降至0.86±0.06、0.74±0.05和0.66±0.07(P<0.05或P<0.01)。结论外源性糖基化终末产物能诱导血管平滑肌细胞上钠/氢交换蛋白1活化,其机制可能与糖基化终末产物激活糖基化终末产物受体,引起丝裂原活化蛋白激酶信号通路活化有关。

    Abstract:

    Aim To evaluate the effect of advanced glycation end products(AGE) on the activity of Na+/H+ exchanger 1(NHE1) of vascular smooth muscle cell(VSMC) in rats. Methods Patch-attaching method was used in primary and continuous culture of the VSMC.The cultured VSMC of rat were exposed to AGE for 24 hours,and the activity of NHE1 was observed by using the fluorescent dye BCECF. Results AGE at concentration 1,5,10 mg/L can concentration-dependently increase the NHE1 activity,from 0.66±0.05 to 0.71±0.03,0.80±0.07,0.88±0.06 respectively;Results of RT-PCR showed that AGE can significantly increase the expression of NHE1 mRNA from 0.77±0.07 to 0.88±0.08,1.23±0.06,1.33±0.08 respectively(P<0.05 or P<0.01);Pretreatment with the inhibitor of the receptor for advanced glycation end products(RAGE) anti-RAGE and inhibitor of mitogen-activated protein kinase(MAPK)PD98059 at concentration 1,10,25 μmol/L can significantly decrease the NHE1 activity from 0.94±0.05 to 0.86±0.06,0.74±0.05,0.66±0.07 respectively compared with the AGE treatment group(P<0.05 or P<0.01). Conclusion AGE can significantly increase the NHE1 activity,and the mechanism may be related to the RAGE activated and the MAPK.
