兔动脉粥样硬化形成过程中血管平滑肌细胞层肉碱棕榈酰转移酶Ⅰ mRNA表达下调

    摘要:

    目的观察高脂饮食复制兔动脉粥样硬化过程中血管平滑肌组织基因表达的变化,以探讨动脉粥样硬化形成是否伴有血管平滑肌细胞能量代谢基因表达的变化。方法36只新西兰大白兔随机分成对照组和高脂组,在饲养的第8、16和第24周用逆转录聚合酶链反应方法检测血管壁平滑肌细胞层肉碱棕榈酰转移酶ⅠmRNA的表达水平。结果高胆固醇饮食诱导兔形成动脉粥样硬化的过程中,16周和24周组同对照组比较肉碱棕榈酰转移酶ⅠmRNA的表达明显减少,24周同16周比较也明显减少。结论兔动脉粥样硬化形成过程中,随时间延长血管平滑肌细胞层肉碱棕榈酰转移酶ⅠmRNA表达下调。

    Abstract:

    Aim To observe the expression of carnitine palmitoyltransferase-Ⅰ(CPT-Ⅰ) gene in vascular smooth muscle tissue during atherosclerosis at 8th,16th,24th week after high-cholesterol diet to rabbit to determine whether atherogenesis is associated with the changes in the gene expression of vascular smooth muscle tissue energy metabolism. Methods 36 rabbits were randomly and equally assigned to control group and high-cholesterol diet group.The gene expression of CPT-Ⅰ were detected in vascular smooth muscle tissue of aorta thoracalis by semi-quantitative reverse transcription polymerase chain reaction(RT-PCR) at the end of 8th,16th,and 24th week. Results 16 and 24 weeks after high-cholesterol diet,the atherosclerosis formed and expression of CPT-Ⅰ mRNA was reduced.Compared with that of 16th week,CPT-Ⅰ mRNA was reduced at the end of 24 th week. Conclusion During atherosclerosis of rabbit,the expression of CPT-Ⅰ mRNA was downregulated in vascular smooth muscle tissue.
