罗格列酮对冠心病患者外周血单核细胞三磷酸腺苷结合盒转运子A1表达的影响

    摘要:

    目的以冠心病患者外周血单核细胞为研究对象,观察不同浓度的罗格列酮对体外培养的单核细胞三磷酸腺苷结合盒转运子A1表达的影响,以探讨罗格列酮对动脉粥样硬化发生发展的影响。方法密度梯度离心法分离冠心病患者外周血单核细胞,再分别与0、0.1、1.0及10.0μmol/L罗格列酮作用24 h,提取各组细胞总RNA,逆转录聚合酶链反应分别检测三磷酸腺苷结合盒转运子A1、过氧化体增殖物激活型受体γ及肝X受体αmRNA的表达,Western blot检测三磷酸腺苷结合盒转运子A1蛋白的表达。结果罗格列酮引起冠心病患者外周血单核细胞三磷酸腺苷结合盒转运子A1 mRNA与蛋白表达明显增加(p<0.05),过氧化体增殖物激活型受体γ和肝X受体αmRNA表达亦上调(p<0.05)。结论罗格列酮可能通过过氧化体增殖物激活型受体γ-肝X受体α-三磷酸腺苷结合盒转运子A1途径影响外周血单核细胞三磷酸腺苷结合盒转运子A1的表达,从而在动脉粥样硬化的防治过程中发挥重要作用。

    Abstract:

    Aim To study the rosiglitazone on the expression of ATP binding cassette transporter A1(ABCA1) mRNA and protein in monocytes of coronary heart disease. Methods Monocytes were isolated by density gradient method from blood of coronary heart disease,and then incubated with 0,0.1,1.0 and 10.0 μmol/L rosiglitazone for 24 h,total RNA of monocytes were abstracted,reverse transcription-polymerase chain reaction(RT-PCR) were performed to inspect the expression of ABCA1,peroxisome proliferators-activated receptor γ(JPPARγ) and liver X receptor α(LXRα) mRNA,Western blot were performed to inspect the expression of ABCA1 protein level. Results Rosiglitazone could increase the expression of ABCA1 mRNA and protein in monocytes(p<0.05).Rosiglitazone increased the expression of JPPARγ and LXRα mRNA in monocytes too(p<0.05). Conclusions Rosiglitazone could likely via JPPARγ-LXRα-ABCA1 pathway increase the expression of ABCA1 mRNA and protein in monocytes.It implied that rosiglitazone might play an important role in the prevention of atherosclerosis by increasing the gene expression of ABCA1.
