    摘要:

    为探讨粒细胞—巨噬细胞集落刺激因子对人内皮细胞基质金属蛋白酶分泌及活性的影响,体外培养人脐静脉内皮细胞,分别应用蛋白印迹法和酶谱法检测基质金属蛋白酶2的含量和活性。结果发现,与对照组相比,不同浓度(5、2 5和5 0 μg/L)粒细胞—巨噬细胞集落刺激因子可使人内皮细胞基质金属蛋白酶2的蛋白表达和活性增强,且随粒细胞—巨噬细胞集落刺激因子剂量增加而增强。提示粒细胞—巨噬细胞集落刺激因子可通过刺激人内皮细胞基质金属蛋白酶2蛋白的表达,并增强其活性,促进内皮细胞对细胞外基质的降解,从而易于动脉粥样硬化斑块内炎性细胞的渗入,内皮细胞的迁移及新血管的形成,从而对斑块的稳定性产生影响

    Abstract:

    Aim To observe the effect of granular-macrophage colony stimulating factor (GM-CSF) on the expression and activity of matrix metalloproteinases (MMP). Methods Human endothelial cells were cultured in vitro and the protein expressions of MMP-2 were detected by Western blot. MMP-2'activity was detected by Zymography. Results The results revealed that the protein expression and activity of MMP-2 were significantly increased after endothelial cells were stimulated by GM-CSF. And the protein expression and activity of MMP-2 were in a dose-dependent and time-dependent manner. Conclusions The results indicate that GM-CSF can obviously increase the protein expression and activity of MMP-2 in human endothelial cells. So GM-CSF could enhance the breakdown of matrix in lesions of atherosclerosis which could probably induce vulnerable plaque.
