    通过3H-TdR掺入实验和RNA印迹分析,观察了内皮素-1对大鼠血管平滑肌细胞DNA合成及细胞增殖相关基因表达的形响。结果表明,内皮素-1作用于血管平滑肌细胞12h,3H-TdR掺入开始增加,24h达到峰值。在内皮素-1作用下,原癌基因c-fos和c-jun进行快速瞬间表达,其表达活性在30min达到峰值,3h后恢复到原来水平;P34cdc2激酶基因在内皮素-1刺激12h后表达活性开始增加,至24h仍维持在较高的水平。本文提示,内皮素-1对血管平滑肌细胞DNA合成的诱导作用与促进细胞增殖相关基因表达有关。

    Abstract:

    The effects of endothelin-1 on DNA synthesis and expression of genes related to cell proliferation in rat vascular smooth muscle cells (VSMC) were investigated by 3H-TdR incorporation and Northern blot analysis. The results suggest that 3H-TdR uptake increased at 12 h after stimulation with endothelin-1, with reaching a peak at 24 h.Endothelin-1 induced rapidly and transiently the expression of the proto-oncogenes c-fos and c-jun,the activity of their expression peaked at 30 min after stimulation, and rapidly declined there after. The expression of the p34cdc2 kinase gene augmented at 12 h after endothelin-1 stimulation and continuously expressed at relatively high levels until 24 h. These data indicate that endothelin-1 stimulates DNA synthesis of VSMC by inducing the transcription of genes related to cell proliferation.
